Regeneration allows organisms to restore lost tissues, and Hydra vulgaris is a powerful model due to its ability to regenerate its entire body from pieces of tissue. Hydra has been found to replenish lost cells through transdifferentiation, where differentiated cells convert into other cells without reverting to a stem cell state. Gland cells, which aid in digestion, are found in both the head and body. Previous studies found body gland cells transdifferentiating into one of the two head gland cell types, but the origin of the other head gland cell remains unknown. I initially hypothesized that body gland cells contribute to the regeneration of this second head gland cell type through transdifferentiation. Using fluorescence in situ hybridization to assess cell marker co- expression, we found no direct evidence of transdifferentiation, suggesting head gland cells may be replenished by interstitial stem cells or an intermediate step exists where the tested markers are not expressed. To test this, we depleted iSCs and observed slower regeneration but no coexpression, indicating a need for longer time points. We aim to identify new markers that could show intermediate cell states. Understanding these mechanisms provides insights into cellular plasticity and may inform regenerative medicine.
