inc Scaffold to Model an Enzyme Active Site New Delhi Metallo-B-lactamase (NDM) is a protein with a di-zinc active site that hydrolyzes carbapenem and penicillin antibiotics. NDM is one route that bacteria use to become resistant to antimicrobials, and therefore NDM inhibitors would be valuable tools for fighting antimicrobial resistant pathogens. We set out to model the NDM active site using a small molecule to examine potential drug binding to the di-zinc core. A ligand scaffold, HL, for supporting two metals was synthesized using a microwave reactor and purified using extraction and crystallization. The microwave reaction was rapid and more efficient than standard synthetic methods. The ligand was then metalated with zinc acetate and sodium tetraphenylborate to generate the complex LZn,(OAc),] BPh,. This dizinc complex is being studied for interaction with potential zinc-binding drugs such as 8-hydroxyquinolin derivatives, to provide insight into how potential drugs interact with two zinc atoms.
